New Links Between Aging, Cancer And Stem Cells

Researchers at the University of Michigan have managed to show that four genes that have previously come under scientific scrutiny as they are believed to help control cancer, also play key roles in the process of aging for humans and in human adult stem cell regulation.

Currently, the biological basis of ageing is not well-understood. There are many theories, each with some evidence supporting it, but no one universally accepted theory of why and how ageing occurs. Diseases which are a product of ageing can also contribute to it, and many of these can be treated with stem cells - for example, there is stem cell therapy for stroke, stem cell therapy for diabetes, and stem cell therapy for Parkinson’s disease.

While the role of stem cells in ageing is seen, but not understood, there also exist several other theories. The telemore theory, where telemores at the end of chromosomes shorten with each successive division and then prevent further multiplication, is one. The free radical theory is another popular one, which states that ageing is a result of the effects of free radicals damaging our cells.

However, stem cell research has shown that human adult stem cells certainly play a large part in the process, or prevention, of ageing.

The four genes are known to scientists as Ink4a, Arf, Hmga2 and Iet-7b. They have been shown to suppress tumor formation, and now are known to also regulate the ability of human adult stem cells to replace worn-out tissues, and regulate the way that adult stem cells shut down during ageing.

These four genes switch themselves on and off, coordinating with each other as cells get older to help reduce the risk of cancer. The internal stem cell therapy function in ageing tissue is shut down, reducing its ability to regenerate.

One of the outcomes of stem cell research in conjunction with cancer research has been to show that mutated cells that are allowed to divide may cause cancer - and cells mutate as they age. Stopping mutated cells from reproducing is your body’s way of helping stop cancer.

He said: “The genes identified in this study work together to reduce the function of adult stem cells as they age. Embryonic stem cells offer the advantage of not aging, not turning on this pathway. If you need to generate large numbers of cells to treat a major public health problem, such as juvenile diabetes this is a big advantage.”

It was the same team that showed two years ago that ink4a, already known as a tumor suppressor, increases in activity as ageing progresses and also shuts down stem cell replication in older mice.

Young Black Men In South Unaware Of H.I.V. Risks, Survey Finds

A small survey of young black men from the South who tested positive for H.I.V. in their teens and early 20s discovered that most had participated in risky sexual behaviors but thought it unlikely they would be infected, according to the federal C.D.C. and Prevention.

More than 50% of the 29 gay or bisexual men researched said they had participated in unprotected anal sex in the year before they were infected and had had sex with slightly older men, the survey found. Both are risky behaviors, yet the vast majority of the young men said they had not thought that they would ever be infected.

Young black gay and bisexual men are becoming infected with H.I.V. at very alarming rates, mostly in the South, and health officials are trying to analyze their risk factors in order to refine education and intervention strategies.

We need to make sure that H.I.V. infection does not become a rite of passage for young black men who are bisexual, said Dr. Alexandra Oster, one of the authors of the survey published last week in the agency’s Morbidity and Mortality Weekly Report.

After the Mississippi State Department of Health notified the C.D.C. in late 2007 that the amount of new H.I.V. diagnoses had spiked at a sexually transmitted disease clinic serving Jackson, Miss., , the agencies teamed up to do the survey. The number of newly diagnosed H.I.V. cases among all black men in the Jackson area had increased 20 percent between 2004-2005 and 2006-2007, but infections among those ages 17 to 25 had jumped 45 percent.

The agencies researched 29 black men ages 17 to 25 who had tested positive for H.I.V. between 2006 and 2008 and who said they had sex with other men. Twenty reported having unprotected anal intercourse with a man during the year before their positive H.I.V. tests, and 16 reported having male sex partners who were 26 or older.

Having older sex partners increases the possibility of infection because older men are usually more likely than younger men to be infected.

Only three of the 29 men thought it likely they would get H.I.V. during their lifetime. More than 50% thought it unlikely or very unlikely, the survey found.

Health officials were more troubled about the lack of routine H.I.V. testing in this group of young men. Six of the men had not had a single H.I.V. test in the two years prior to testing positive, and five had had only one test in the two years before the positive result, the survey found.

These men might have taken what they believed were reasonable steps to decrease their risk, but unfortunately the rates of H.I.V. infection are so high in this population that in some cases people have partners who are H.I.V. positive and do not know it, said Richard Wolitski, director of the C.D.C.’s division of H.I.V./AIDS Prevention.
